With Sanger sequencing technology, we can cover up to 1200 bp of your samples, starting either from purified or unpurified PCR products, or plasmids. If needed, we can also perform gDNA extraction to fit your project.

Method

The simplicity of Sanger sequencing is what makes it so fast and cost-effective. The technology uses already established lab protocols such as PCR and gel electrophoresis along with some critical additions to sequence the DNA fragments. Sanger sequencing is also known as the chain-termination method which describes the chain-terminating nucleotides that are added during the PCR step. These are known as ddNTPs and are added in the PCR reaction mix along with dNTPs so that the fragments still elongate as usual. The ddNTPs are vital for this method of sequencing as they stop the elongation step of the PCR reaction, resulting in DNA fragments of different sizes.

The DNA fragments are then separated on a gel which results in a laddered appearance because of their varying sizes. The ddNTPs have four differently coloured fluorophores attached which, when scanned by a laser, signals the type of base that it is bound to. Once the series of bases has been established, you can build your sequence together and now know the complete sequence of your fragment.
